Microscopic and flowcytometric detection of immunofluorescent OPN staining showed strong surface expression on virtually all DTC, whereas perinuclear intracellular expression and low surface expression was observed in subsets of PTC. A single OPN transcript of 1.6 kb was found in PTC and DTC. Both conditioned medium and cell lysates of PTC and DTC contained different OPN MW forms, indicating the presence of posttranslational modifications. Consequently, no differences were found between PTC and DTC in terms of OPN production. In addition, no co-localization was found between intracellular OPN and endocytosed dextran in PTC. We demonstrated production and secretion of OPN by primary cultures of human PTC as well as DTC. Human OPN expression pattern observed in vitro was in agreement with expression pattern seen after renal injury in the rat and could not be explained by differences in OPN production or endocytosis by PTC.
